Ubuntu 20.04 64bit on raspi 4 howto transfer to USB SSD

Hi I have started with a Raspi 4b 8Gb.
It is now running Ubuntu 20.04 with mate from a microsd-card.
I want to run it from an USB3-SSD.
The first action is to transfer everything from the microsd card to the USB-SSD.
Just how do i do that?
I have a dualboot kubuntu 18.04 / Windows 10 pro Laptop that i can use but I am unsure how to transfer the whole system to usb-SSD.
I never did anything like that before, what is the easiest way? Can I use ext4 as filesystem on the SSD, or must the bootpartition be fat?
