News of McCaffrey's injury arrives three days before the 49ers are scheduled to open their season Sunday against the Seattle Seahawks.
McCaffrey was limited to just four games last season while dealing with calf and Achilles injuries.
When healthy, McCaffrey is one of the NFL's most potent offensive weapons and an elite dual-threat weapon out of the backfield. Six plays later, Javonte Williams ran for a one-yard touchdown to give the Cowboys a 7-0 lead.
